The role of proteasome inhibitor MG132 in cisplatin resistant ovarian cancer

Platinum based combined chemotherapy have been proved to be the most effective drugs for ovarian cancer treatment, but it is difficult treat cisplatin resistant cancer. Carbobenzoxy-L-leucy-L-Leucyl-L-Leucinal (MG132) a reversible tripeptide aldehyde proteasome inhibitor, purpose of this study was observe effect MG132 on SKOV3 cell and OVCAR-3 expression autophagy apoptosis related factors. The cells were divided into four groups: control, MG132, cisplatin, combination groups. Cell growth detected by counting kit-8 (CCK-8) assay. apoptotic rates cycle flow cytometer (FCM). Beclin1, Light chain 3 (LC3) Caspase3 western blotting reverse transcription-polymerase reaction (RT-PCR). Detection bodies 4,6-Diamidino-2-phenylindole dihydrochloride (DAPI) staining. CCK-8 assay demonstrated that survival rate in groups lower than monotherapy group. FCM showed higher group (p < 0.05). Western RT-PCR LC3 DAPI staining production In conclusion, can inhibit cells, its inhibitory autophagy, expected synergistic antitumor with cisplatin.
